Abstract A mutualistic co‐evolution exists between the host and its associated microbiota in the human body. Bacteria establish ecological niches in various tissues of the body, locally influencing their physiology and functions, but also contributing to the well‐being of the whole organism through systemic communication with other distant niches (axis). Emerging evidence indicates that when the composition of the microbiota inhabiting the niche changes toward a pathogenic state (dysbiosis) and interactions with the host become unbalanced, diseases may present. In addition, imbalances within a single niche can cause dysbiosis in distant organs. Current research efforts are focused on elucidating the mechanisms leading to dysbiosis, with the goal of restoring tissue homeostasis. In vitro models can provide critical experimental platforms to address this need, by reproducing the niche cyto‐architecture and physiology with high fidelity. This review surveys current in in vitro host–microbiota research strategies and provides a roadmap that can guide the field in further developing physiologically relevant in vitro models of ecological niches, thus enabling investigation of the role of the microbiota in human health and diseases. Lastly, given the Food and Drug Administration Modernization Act 2.0, this review highlights emerging in vitro strategies to support the development and validation of new therapies on the market.more » « lessFree, publicly-accessible full text available May 14, 2026

Aims. Globular clusters (GCs) are known to host distinct stellar populations, characterized by different chemical compositions. Despite extensive research, the origin of these populations remains elusive. According to many formation scenarios, the second population (2P) originated within a compact and denser region embedded in a more extended first population (1P) system. As a result, 2P binaries should be disrupted at a larger rate than 1P binaries. For this reason, binary systems offer valuable insight into the environments in which these stellar populations formed and evolved. Methods. We analyzed the fraction of binaries among 1P and 2P M dwarfs in the outer region of NGC 288 using Hubble Space Telescope data. We combined our results with those from a previous work, where we inferred the fraction of 1P and 2P binaries in the cluster center. Results. In the outer region, we find a predominance of 1P binaries (97−3+1%) compared to 2P binaries (3 ± 1%) corresponding to an incidence of binaries with a mass ratio (i.e., the ratio between the masses of the primary and secondary star) greater than 0.5 equal to 6.4 ± 1.7% for the 1P and 0.3 ± 0.2% for the 2P. These binary fractions and incidences differ from those found in the cluster’s central region, where the 1P and 2P exhibit similar binary incidences and fractions. Abstract Guided by concepts from life history (LH) theory, a large human research literature has tested the hypothesis that exposures to extrinsic mortality (EM) promote the development of faster LH strategies (e.g., earlier/faster reproduction, higher offspring number). A competing model proposes that, because EM in the past was intimately linked to energetic constraints, such exposures specifically led to the development of slower LH strategies. We empirically address this debate by examining (1) LH variation among small-scale societies under different environmental conditions; (2) country-, regional- and community-level correlations between ecological conditions, mortality, maturational timing, and fertility; (3) individual-level correlations between this same set of factors; and (4) natural experiments leveraging the impact of externally-caused changes in mortality on LH traits. Partially supporting each model, we found that harsh conditions encompassing energetic stress and ambient cues to EM (external cues received through sensory systems) havecountervailing effectson the development of LH strategies, both delaying pubertal maturation and promoting an accelerated pace of reproduction and higher offspring number. We conclude that, although energetics are fundamental to many developmental processes, providing afirst tierof environmental influence, this first tier alone cannot explain these countervailing effects. An importantsecond tierof environmental influence is afforded by ambient cues to EM. We advance a 2-tiered model that delineates this second tier and its central role in regulating development of LH strategies. Consideration of the first and second tier together is necessary to account for the observed countervailing shifts toward both slower and faster LH traits.more » « less

Almost all globular clusters (GCs) contain multiple stellar populations consisting of stars with varying helium and light-element abundances. These populations include first-population stars, which exhibit similar chemical compositions as halo-field stars with comparable [Fe/H], and second-population stars, characterized by higher helium and nitrogen abundances along with reduced levels of oxygen and carbon. Nowadays, one of the most intriguing open questions about GCs pertains to the formation and evolution of their multiple populations. Recent works based on N-body simulations of GCs show that the fractions and characteristics of binary stars can serve as dynamic indicators of the formation period of multiple-population GCs and their subsequent dynamical evolution. Nevertheless, the incidence of binaries among multiple populations is still poorly studied. Moreover, the few available observational studies focus only on the bright stars of a few GCs. We used deep images of the GC 47 Tucanae collected with theJames Webband theHubblespace telescopes to investigate the incidence of binaries among multiple populations of M dwarfs and bright main- sequence stars. To reach this objective, we used UV, optical, and near-infrared filters to construct photometric diagrams that allowed us to disentangle binary systems and multiple populations. Moreover, we compared these observations with a large sample of simulated binaries. In the cluster central regions, the incidence of binaries among first-population stars is only slightly higher than that of second- population stars. In contrast, in the external regions, the majority of the studied binaries (≳85%) are composed of first-population stars. These results are consistent with the GC formation scenarios in which the second-population stars originate in the cluster’s central region, forming a compact and dense stellar group within a more extended system of first-population stars.more » « lessFree, publicly-accessible full text available June 1, 2026

In contrast to fairly good knowledge of dissolved carbon and major elements in great Arctic rivers, seasonally resolved concentrations of many trace elements remain poorly characterized, hindering assessment of the current status and possible future changes in the hydrochemistry of the Eurasian Arctic. To fill this gap, here we present results for a broad suite of trace elements in the largest rivers of the Russian Arctic (Ob, Yenisey, Lena, and Kolyma). For context, we also present results for major elements that are more routinely measured in these rivers. Water samples for this study were collected during an international campaign called PARTNERS from 2004 through 2006. A comparison of element concentrations obtained for Arctic rivers in this study with average concentrations in the world’s rivers shows that most elements in the Arctic rivers are similar to or significantly lower than the world average. The mineral content of the three greatest rivers (Ob, Yenisey, and Lena) varies within a narrow range (from 107 mg/L for Yenisey to 123 mg/L for Ob). The Kolyma’s mineral content is significantly lower (52.4 mg/L). Fluxes of all major and trace elements were calculated using average concentrations and average water discharge for the 2004–2006 period. Based on these flux estimates, specific export (i.e., t/km2/y) for most of the elements was greatest for the Lena, followed by the Yenisey, Ob, and Kolyma in decreasing order. Element pairwise correlation analysis identified several distinct groups of elements depending on their sources and relative mobility in the river water. There was a negative correlation between Fe and DOC concentration in the Ob River, which could be linked to different sources of these components in this river. The annual yields of major and trace elements calculated for each river were generally consistent with values assessed for other mid-size and small rivers of the Eurasian subarctic.more » « less

This study aimed to evaluate the impact of Nystatin oral rinse on salivary and supragingival microbiota in adults with oral candidiasis and identify predictive factors related to individuals’ responses to Nystatin. The trial involved twenty participants who used 600,000 International Units/application of Nystatin oral rinse for seven days, four times a day, and were followed up at one week and three months after the rinse. The salivary and plaque microbiome of the participants were assessed via 16S rDNA amplicon sequencing. Overall, salivary and plaque microbiomes remained stable. However, among the participants (53 percent) who responded to Nystatin rinse (defined as free of oral Candida albicans post treatment), Veillonella emerged as a core genus alongside Streptococcus and Actinomyces in supragingival plaque at the 3-month follow-up. Furthermore, statistical models were fit to identify predictive factors of Nystatin rinse success (elimination of C. albicans) or failure (remaining C. albicans). The results revealed that an increased level of salivary Interferon (IFN)-γ-inducible protein (IP-10), also known as C-X-C motif chemokine ligand 10 (CXCL10), was an indicator of a failure of responding to Nystatin rinse. Future clinical trials are warranted to comprehensively assess the impact of antifungal treatment on the oral flora.more » « less

Small populations with limited range are often threatened by inbreeding and reduced genetic diversity, which can reduce fitness and exacerbate population decline. One of the most extreme natural examples is the Devils Hole pupfish ( Cyprinodon diabolis ), an iconic and critically endangered species with the smallest known range of any vertebrate. This species has experienced severe declines in population size over the last 30 years and suffered major bottlenecks in 2007 and 2013, when the population shrunk to 38 and 35 individuals, respectively. Here, we analysed 30 resequenced genomes of desert pupfishes from Death Valley, Ash Meadows and surrounding areas to examine the genomic consequences of small population size. We found extremely high levels of inbreeding ( F ROH = 0.34–0.81) and an increased amount of potentially deleterious genetic variation in the Devils Hole pupfish as compared to other species, including unique, fixed loss-of-function alleles and deletions in genes associated with sperm motility and hypoxia. Additionally, we successfully resequenced a formalin-fixed museum specimen from 1980 and found that the population was already highly inbred prior to recent known bottlenecks. We thus document severe inbreeding and increased mutation load in the Devils Hole pupfish and identify candidate deleterious variants to inform management of this conservation icon.more » « less

Early childhood caries (ECC) is a disease that globally affects pre-school children. It is important to identify both protective and risk factors associated with this disease. This paper examined a set of saliva samples of Thai mother–child dyads and aimed to analyze how the maternal factors and oral microbiome of the dyads influence the development of ECC. However, heterogeneous latent subpopulations may exist that have different characteristics in terms of caries development. Therefore, we introduce a novel method to cluster the correlated outcomes of dependent observations while selecting influential independent variables to unearth latent groupings within this dataset and reveal their association in each group. This paper describes the discovery of three heterogeneous clusters in the dataset, each with its own unique mother–child outcome trend, as well as identifying several microbial factors that contribute to ECC. Significantly, the three identified clusters represent three typical clinical conditions in which mother–child dyads have typical (cluster 1), high–low (cluster 2), and low–high caries experiences (cluster 3) compared to the overall trend of mother–child caries status. Intriguingly, the variables identified as the driving attributes of each cluster, including specific taxa, have the potential to be used in the future as caries preventive measures.more » « less

Abstract Hydrogel-encapsulated catalysts are an attractive tool for low-cost intensification of (bio)-processes. Polyvinyl alcohol-sodium alginate hydrogels crosslinked with boric acid and post-cured with sulfate (PVA-SA-BS) have been applied in bioproduction and water treatment processes, but the low pH required for crosslinking may negatively affect biocatalyst functionality. Here, we investigate how crosslinking pH (3, 4, and 5) and time (1, 2, and 8 h) affect the physicochemical, elastic, and process properties of PVA-SA-BS beads. Overall, bead properties were most affected by crosslinking pH. Beads produced at pH 3 and 4 were smaller and contained larger internal cavities, while optical coherence tomography suggested polymer cross-linking density was higher. Optical coherence elastography revealed PVA-SA-BS beads produced at pH 3 and 4 were stiffer than pH 5 beads. Dextran Blue release showed that pH 3-produced beads enabled higher diffusion rates and were more porous. Last, over a 28-day incubation, pH 3 and 4 beads lost more microspheres (as cell proxies) than beads produced at pH 5, while the latter released more polymer material. Overall, this study provides a path forward to tailor PVA-SA-BS hydrogel bead properties towards a broad range of applications, such as chemical, enzymatic, and microbially catalyzed (bio)-processes.more » « less
